Title: BOIN - a Novel Platform for Designing Early Phase Clinical Trials

Abstract
Bayesian optimal interval (BOIN) designs belong to a new class of early phase trial designs known as model-assisted designs, which utilize statistical models for efficient decision making, but their dose transition rules can be pre-tabulated as the 3+3 design. The BOIN design can be implemented in a simple way as the 3+3 design, but yields superior performance comparable to model-based designs. In this talk, we will introduce a suite of BOIN designs that can be used to find the maximum tolerated dose (MTD) for single-agent trials, drug-combination trials, as well as trials with late-onset toxicity, under a unified framework. The freely available software at www.trialdesign.org will be demonstrated using trial examples.
